    Cell surface-specific N-glycan profiling in breast cancer

    Aberrant changes in specific glycans have been shown to be associated with immunosurveillance, tumorigenesis, tumor progression and metastasis. In this study, the N-glycan profiling of membrane proteins from human breast cancer cell lines and tissues was detected using modified DNA sequencer-assisted fluorophore-assisted carbohydrate electrophoresis (DSA-FACE). The N-glycan profiles of membrane proteins were analyzed from 7 breast cancer cell lines and MCF 10A, as well as from 100 pairs of breast cancer and corresponding adjacent tissues. The results showed that, compared with the matched adjacent normal tissue samples, two biantennary N-glycans (NA2 and NA2FB) were significantly decreased (p <0.0001) in the breast cancer tissue samples, while the triantennary glycan (NA3FB) and a high-mannose glycan (M8) were dramatically increased (p = 0.001 and p <0.0001, respectively). Moreover, the alterations in these specific N-glycans occurred through the oncogenesis and progression of breast cancer. These results suggested that the modified method based on DSA-FACE is a high-throughput detection technology that is suited for analyzing cell surface N-glycans. These cell surface-specific N-glycans may be helpful in recognizing the mechanisms of tumor cell immunologic escape and could be potential targets for new breast cancer drugs

    Pressure-induced superconductivity in charge-density-wave compound LaTe2-xSbx (x=0 and 0.4)

    Here, we have grown single crystals of LaTe2-xSbx (x=0 and 0.4) with continuously adjustable CDW. High-pressure x-ray diffraction show LaTe2 does not undergo phase transition and keep robust below 40 GPa. In-situ high-pressure electrical measurements show LaTe2-xSbx undergo semiconductor-metal-superconductivity transition at 4.6 and 2.5 GPa, respectively. With the doping of Sb, the highest Tc increases from 4.6 to 6.5 K. Theoretical calculations reveal that the CDW has been completely suppressed and the calculated Tc is about 2.97 K at 4.5 GPa, consistent with the measured value. Then, the pressure-induced superconductivity in LaTe2-xSbx can be explained in the framework of the BCS theory.Comment: 11 pages, 5 figure

    Isolation and complete genomic characterization of H1N1 subtype swine influenza viruses in southern China through the 2009 pandemic

    <p>Abstract</p> <p>Background</p> <p>The swine influenza (SI) is an infectious disease of swine and human. The novel swine-origin influenza A (H1N1) that emerged from April 2009 in Mexico spread rapidly and caused a human pandemic globally. To determine whether the tremendous virus had existed in or transmitted to pigs in southern China, eight H1N1 influenza strains were identified from pigs of Guangdong province during 2008-2009.</p> <p>Results</p> <p>Based on the homology and phylogenetic analyses of the nucleotide sequences of each gene segments, the isolates were confirmed to belong to the classical SI group, with HA, NP and NS most similar to 2009 human-like H1N1 influenza virus lineages. All of the eight strains were low pathogenic influenza viruses, had the same host range, and not sensitive to class of antiviral drugs.</p> <p>Conclusions</p> <p>This study provides the evidence that there is no 2009 H1N1-like virus emerged in southern China, but the importance of swine influenza virus surveillance in China should be given a high priority.</p

    Alteration of liver N-glycome in patients with hepatocellular carcinoma

    Purpose: Alteration of liver function during progression of hepatocellular carcinoma (HCC) and cirrhosis affects the serum glycoprotein pattern. In this study, the changes in the N-glycome in liver tissue from patients with hepatocellular carcinoma and cirrhosis caused by hepatitis B virus infection were investigated to find out the relationship between this maker and liver disease. Methods: Twenty patients, 11 with cirrhosis and 9 with hepatocellular carcinoma, and 15 healthy donors were involved in this study. Liver protein N-glycans were profiled using the DSA-FACE technique developed in our laboratory. To further analyze the fucosylation status of these liver glycans Western lectin blots of total liver proteins were performed using Aspergillus oryzae lectin (AOL) as probe, which is a carbohydrate-binding protein that recognizes specifically α-1,6-fucosylated glycans. Results: The N-glycome of liver proteins in patients with HBV related HCC and cirrhosis was analyzed. Compared with healthy donors, the N-glycome had significantly less (p < 0.05) high mannose (M8) in both groups of patients. The total core α-1,6-fucosylation in total liver glycopro-teins was dramatically increased during the progress of hepatocellular carcinoma and cirrhosis compared to the controls. Conclusion: These results show that fucosylation not only increases in serum proteins but also in liver tissue itself of patients with HBV related HCC and cirrhosis

    miR-24 is involved in vertebrate LC-PUFA biosynthesis as demonstrated in marine teleost Siganus canaliculatus

    Recently, microRNAs (miRNAs) have emerged as crucial regulators of lipid metabolism. However, the miRNA-mediated regulatory mechanism on long-chain (≥C20) polyunsaturated fatty acids (LC-PUFA) biosynthesis in vertebrates remains largely unknown. Here, we address a potentially important role of miRNA-24 (miR-24) in the regulation of LC-PUFA biosynthesis in rabbitfish Siganus canaliculatus. miR-24 showed significantly higher abundance in liver of rabbitfish reared in brackish water than in seawater for fish fed vegetable oil diets and in S. canaliculatus hepatocyte line (SCHL) cells incubated with alpha-linolenic acid (ALA) than the control group. Similar expression patterns were also observed on the expression of sterol regulatory element-binding protein-1 (srebp1) and LC-PUFA biosynthesis related genes. While opposite results were observed on the expression of insulin-induced gene 1 (insig1), an endoplasmic reticulum membrane protein blocking Srebp1 proteolytic activation. Luciferase reporter assays revealed rabbitfish insig1 as a target of miR-24. Knockdown of miR-24 in SCHL cells resulted in increased Insig1 protein, and subsequently reduced mature Srebp1 protein and expression of genes required for LC-PUFA biosynthesis, and these effects could be attenuated after additional insig1 knockdown. Opposite results were observed with overexpression of miR-24. Moreover, increasing endogenous insig1 by knockdown of miR-24 inhibited Srebp1 processing and consequently suppressed LC-PUFA biosynthesis in rabbitfish hepatocytes. These results indicate a potentially critical role for miR-24 in regulating LC-PUFA biosynthesis through the Insig1/Srebp1 pathway by targeting insig1. This is the first report of miR-24 involved in LC-PUFA biosynthesis and thus may provide knowledge on the regulatory mechanisms of LC-PUFA biosynthesis in vertebrates

    Robust anomalous Hall effect in ferromagnetic metal under high pressure

    Recently, the giant intrinsic anomalous Hall effect (AHE) has been observed in the materials with kagome lattice. In this study, we systematically investigate the influence of high pressure on the AHE in the ferromagnet LiMn6Sn6 with clean Mn kagome lattice. Our in-situ high-pressure Raman spectroscopy indicates that the crystal structure of LiMn6Sn6 maintains a hexagonal phase under high pressures up to 8.51 GPa. The anomalous Hall conductivity (AHC) {\sigma}xyA remains around 150 {\Omega}-1 cm-1, dominated by the intrinsic mechanism. Combined with theoretical calculations, our results indicate that the stable AHE under pressure in LiMn6Sn6 originates from the robust electronic and magnetic structure.Comment: 11 pages 5 figure

    Pressure-tunable magnetic topological phases in magnetic topological insulator MnSb4Te7

    Magnetic topological insulators, possessing both magnetic order and topological electronic structure, provides an excellent platform to research unusual physical properties. Here, we report a high-pressure study on the anomalous Hall effect of magnetic TI MnSb4Te7 through transports measurements combined with first-principle theoretical calculations. We discover that the ground state of MnSb4Te7 experiences a magnetic phase transition from the A-type antiferromagnetic state to ferromagnetic dominating state at 3.78 GPa, although its crystal sustains a rhombohedral phase under high pressures up to 8 GPa. The anomalous Hall conductance {\sigma}xyA keeps around 10 {\Omega}-1 cm-1, dominated by the intrinsic mechanism even after the magnetic phase transition. The results shed light on the intriguing magnetism in MnSb4Te7 and pave the way for further studies of the relationship between topology and magnetism in topological materials.Comment: 10 pages, 4 figure

    Pressure-induced Superconductivity and Structure Phase Transition in SnAs-based Zintl Compound SrSn2As2

    Layered SnAs-based Zintl compounds exhibit a distinctive electronic structure, igniting extensive research efforts in areas of superconductivity, topological insulators and quantum magnetism. In this paper, we systematically investigate the crystal structures and electronic properties of the Zintl compound SrSn2As2 under high-pressure. At approximately 20.8 GPa, pressure-induced superconductivity is observed in SrSn2As2 with a characteristic dome-like evolution of Tc. Theoretical calculations together with high pressure synchrotron X-ray diffraction and Raman spectroscopy have identified that SrSn2As2 undergoes a structural transformation from a trigonal to a monoclinic structure. Beyond 28.3 GPa, the superconducting transition temperature is suppressed due to a reduction of the density of state at the Fermi level. The discovery of pressure-induced superconductivity, accompanied by structural transitions in SrSn2As2, greatly expands the physical properties of layered SnAs-based compounds and provides a new ground states upon compression.Comment: 15 pages, 6 figures. arXiv admin note: text overlap with arXiv:2307.1562
